Changes in bone density and arthritis score after intervention with metformin in ankylosing spondylitis model mice

二甲双胍治疗强直性脊柱炎模型小鼠后的骨密度与关节炎评分变化

Abstract:

Objective To investigate the effects of metformin on bone mineral density and peripheral arthritis score in ankylosing spondylitis (AS) model mice. Methods The study was from September 2021 to June 2022. The BLAB/c mice were induced by proteoglycan to establish AS mouse models. The mice were divided into an experimental model group, a model control group, and a blank control group, with 10 in each group. The mice in the experimental model group were given metformin, the other mice were given normal saline by gavage once a day for three weeks. The climbing and suspension activities of the mice were observed; the scores of diseased joints were recorded; the bone density was measured in each group. t test was used for the statistical analysis. Results After 8 weeks of induced immunity, the mice showed symptoms of peripheral arthritis, and the AS model mice reached their peak in the arthritis score at 12 weeks. The arthritis score of the mice treated with metformin was significantly lower than that of the model control group. The climbing and suspension activity of the model mice decreased. The bone mineral density of the model group was significantly lower than that of the blank control group. The bone density of the model group was significantly lower than that of the blank group. The bone mineral densities of the model mice treated with metformin were significantly higher than those in the model control group [femur: (0.101±0.003) g/cm2 vs. (0.029±0.009) g/cm2; tibia: (0.070±0.005) g/cm2 vs. (0.025±0.007) g/cm2; lumbar vertebra: (0.060±0.007) g/cm2 vs. (0.043±0.004) g/cm2], with statistical differences (all P<0.05). Conclusion Metformin can significantly improve the symptoms of peripheral arthritis and increase bone mineral density in AS model mice.

摘要:

目的 探讨二甲双胍治疗强直性脊柱炎(ankylosing spondylitis,AS)模型小鼠后的骨密度与外周关节炎评分影响。方法 研究时间为2021年9月至2022年6月。蛋白聚糖诱导BLAB/c小鼠,建立AS小鼠模型,小鼠分为3组:二甲双胍模型组、模型对照组、空白对照组,每组10只。实验模型组小鼠灌胃二甲双胍、模型对照组和空白对照组小鼠灌胃生理盐水,每天1次,连续3周。观察小鼠攀爬悬吊活动情况,病变关节进行评分,测定各组小鼠骨密度。采用t检验进行统计分析。结果 诱导免疫8周后小鼠出现外周关节炎症状,模型小鼠在12周关节炎评分分值达到顶峰。模型小鼠攀爬悬吊活动能力下降。二甲双胍模型组小鼠关节炎评分明显低于模型对照组小鼠,模型对照组小鼠骨密度明显低于空白组的骨密度。二甲双胍模型组小鼠骨密度明显改善,与模型对照组[股骨:(0.101±0.003)g/cm2比(0.029±0.009)g/cm2,胫骨:(0.070±0.005)g/cm2比(0.025±0.007)g/cm2,腰椎:(0.060±0.007)g/cm2比(0.043±0.004)g/cm2]比较,差异均有统计学意义(均P<0.05)。结论 二甲双胍能明显改善AS模型小鼠外周关节炎症状和提升骨密度。
