Study on quality standard of carbonized Scutellariae Radix

黄芩炭饮片质量标准研究

Abstract:

Objective To establish the quality standard of carbonized Scutellariae Radix and provide reference for its quality control. Methods The samples of carbonized Scutellariae Radix were selected for character observation and thin layer chromatography (TLC) identification. The water content, total ashcontent, and alcohol-soluble extract were determined according to the general rules of Chinese Pharmacopoeia (2020 Edition). The content of baicalin in the samples was detected by high performance liquid chromatography (HPLC), and the specific chromatogram was established. Results Carbonized Scutellariae Radix was round or irregularly shaped segment or piece, one side black brown, inside yellow brown, crisp quality. It had a burnt smell and a bitter taste. In the TLC diagram of test samples, fluorescence spots of the same color were shown on corresponding positions of the chromatographs of the control products (baicalein and wogonin). In 10 batches of carbonized Scutellariae Radix, the water content, total ashcontent, and alcohol-soluble extracts were 2.60%-7.36%, 5.03%-8.92%, and 23.08%-48.92%, respectively, with the average values of 5.20%, 6.39%, and 32.84%, respectively. The content of baicalin ranged from 1.61% to 4.33%, with an average of 2.67%. Conclusions In this study, the water content, total ashcontent, alcohol-soluble extract, and specific chromatogram of carbonized Scutellariae Radix were added to the existing quality standards. The minimum limits of water content, total ashcontent, and alcohol-soluble extract in carbonized Scutellariae Radix were determined. Three characteristic peaks should be detected, which were consistent with the retention time of reference peaks of baicalin, baicalein, and wogonin, respectively. The established quality standard could provide reference for the quality control of carbonized Scutellariae Radix.

摘要:

目的 建立黄芩炭的质量标准,为其质量控制提供参考。方法 取黄芩炭样品,进行性状观察、薄层色谱(TLC)鉴别,并按《中国药典(2020年版)》通则方法测定其中水分、总灰分、醇溶性浸出物,利用高效液相色谱法检测其中的黄芩素含量,并建立黄芩炭的特征图谱。结果 黄芩炭为类圆形或不规则形段或片,片面黑褐色,内部黄褐色,质松脆,有焦糊气,味苦。供试品TLC图中,在对照品(黄芩素、汉黄芩素)以及对照药材相对应位置上,显示相同颜色的荧光斑点。10批黄芩炭中,水分、总灰分、醇溶性浸出物分别为2.60%~7.36%、5.03%~8.92%、23.08%~48.92%,平均值分别为5.20%、6.39%、32.84%;黄芩素含量为1.61%~4.33%,平均值为2.67%。结论 本研究在黄芩炭已有质量标准的基础上增加了水分、总灰分、醇溶性浸出物、特征图谱等检测项,拟定了黄芩炭中水分、总灰分、醇溶性浸出物的最低限量,黄芩炭样品应检出3个特征峰,分别与黄芩苷、黄芩素、汉黄芩素对照品参照物峰保留时间一致。所建质量标准可为黄芩炭饮片的质量控制提供参考。
