Effects of Acorus tatarinowii, Polygala tenuifolia, and their compatibility on ox-LDL-induced injury of human umbilical vein endothelial cells

石菖蒲、远志及其配伍对ox-LDL诱导人脐静脉内皮细胞损伤的影响

Abstract: Objective To investigate the effects of Acorus tatarinowii, Polygala tenuifolia, and their compatibility on ox-LDL-induced injury of human umbilical vein endothelial cells (HUVECs). Methods From January to August 2021, HUVECs were cultured, and were randomly divided into a blank group, a model group, and sample groups. The different concentrations (5, 10, and 20 mg/ml) of Acorus tatarinowii, Polygala tenuifolia, and their compatibility were added into the sample groups. The HUVECs proliferation, relative viability test, apoptosis, cell cycle, and expressions of apoptotic proteins Bax and Bcl-2 were detected. One-way ANOVA was applied. Results Acorus tatarinowii, Polygala tenuifolia, and their ompatibility could reduce the apoptosis of HUVECs [the blank group (0.338±0.001), the model group (4.319±0.124), the Acorus tatarinowii group (2.694±0.561), the Polygala tenuifolia group (2.278±0.914), and the Acorus tatarinowii + Polygala tenuifolia group (2.250±0.699)], and increase HUVECs cycle (S+G2)%. Both Acorus tatarinowii and Polygala tenuifolia could down-regulate the apoptotic protein Bax and inhibit the up-regulation of the apoptotic protein Bcl-2, and their compatibility was better than Acorus tatarinowii and Polygala tenuifolia. Conclusion Acorus tatarinowii, Polygala tenuifolia, and their ompatibility can inhibit ox-LDL induced HUVECs injury, and their compatibility is stronger than single Chinese medicine.

摘要: 目的 观察石菖蒲、远志及其配伍对氧化型低密度脂蛋白(ox-LDL)诱导人脐静脉内皮细胞(HUVECs)损伤情况的影响。方法 2021年1月至8月,培养HUVECs,随机分为空白组、模型组和样品组,样品组中分别加入不同浓度(5、10、20 mg/ml)的石菖蒲、远志或石菖蒲+远志,对HUVECs增殖、相对活率试验、细胞凋亡、细胞周期、凋亡蛋白Bax和Bcl-2表达进行检测。采用单因素方差分析。结果 石菖蒲、远志及其配伍均能减少HUVECs的凋亡[空白组(0.338±0.001),模型组(4.319±0.124),石菖蒲组(2.694±0.561),远志组(2.278±0.914),石菖蒲+远志组(2.250±0.699)];石菖蒲、远志及其配伍均能增加HUVECs周期(S+G2)%;远志、石菖蒲均能使凋亡相关蛋白Bax下调,同时抑制凋亡相关蛋白Bcl-2上调,远志+石菖蒲配伍下对Bax及Bcl-2蛋白的影响优于单味中药石菖蒲、远志。结论 石菖蒲、远志及其配伍均能抑制ox-LDL诱导HUVECs损伤,其配伍作用强于单味中药。
