野葛花不同发育期射干苷和染料木素含量经时对比研究

Abstract:

Objective To determine the ongoing change characteristics of the contents of tectoridin and genistein in Puerariae Lobata (Willd.) Ohwio at different development stages (floral bud and flower), so as to provide reference for its rational harvesting and application. Methods The floral bud and flower parts of Puerariae Lobata (Willd.) Ohwio during different harvesting periods were taken as the research samples. The contents of tectoridin and genistein were determined by high performance liquid chromatography (HPLC). The determination was performed on Inertsil C18 column (250 mm×4.6 mm, 5 µm) with mobile phase consisted of acetonitrile - 0.1% phosphoric acid solution (15:85) at the flow rate of 1.0 ml·min-1. The column temperature was 40 ℃ with the detection wavelength as 265 nm, and the sample size was 10 µl. Results The calibration curves of tectoridin and genistein were linear within a range of 0.8-52.0 mg/L (r=1.000 0) and 0.7-44.0 mg/L (r=1.000 0). The average recovery was 98.56% [relative standard deviation (RSD) = 1.97%] and 101.08% (RSD = 1.76%), respectively. The contents of tectoridin in the floral bud and flower samples of Puerariae Lobata (Willd.) Ohwio at different harvesting times showed flower bud > flower, and the content of genistein was flower bud < flower. The highest contents of tectoridin and genistein were from the samples harvested after the beginning of autumn. Conclusion The best harvesting time of Puerariae Lobata (Willd.) Ohwio is after the beginning of autumn.

摘要:

目的 阐明野葛花不同发育期(花蕾、花)射干苷、染料木素含量的经时变化特征,为该药材的合理采收、资源利用提供参考。方法 分别以不同发育期野葛花花、花蕾为样品。采用高效液相色谱法测定射干苷、染料木素的含量,Inertsil C18柱(250 mm×4.6 mm,5 µm),流动相为乙腈-0.1%磷酸水(15∶85),流速为1.0 ml·min-1,柱温40 ℃,检测波长265 nm,进样量为10 µl。结果 射干苷和染料木素的线性范围分别为0.8~52.0 mg/L、0.7~44.0 mg/L,r均为1.000 0,平均加样回收率为98.56%[相对标准偏差(RSD)=1.97%]、101.08%(RSD=1.76%)。不同采收期的野葛花花蕾和花样品中射干苷的含量花蕾>花,染料木素的含量花蕾<花,射干苷和染料木素的含量最高点均为立秋后样品。结论 野葛花立秋后采收较优。
