A novel prognostic feature of lung squamous carcinoma based on cuproptosis associated lncRNAs

基于铜死亡相关lncRNAs构建一种新的肺鳞癌预后特征

Abstract:

Objective To establish a risk prognostic model of cuproptosis associated lncRNAs in lung squamous carcinoma (LUSC) based on the Cancer Genome Atlas (TCGA) database. Methods The gene and clinical data were obtained from the TCGA database. The cuproptosis related genes were obtained from the cuproptosis related studies. The prognosis model of lncRNAs was established by co-expression analysis and Cox regression analysis. The prognostic value of the model was evaluated. The relationship between cuproptosis associated lncRNAs and LUSC was analyzed. The differences between the groups were analyzed by Student's t and Wilcoxon tests. The Kaplan-Meier survival analysis were tested by Log-Rank. Results A total of 5 lncRNAs participated in the construction of the model. The validation analysis found poor overall survival (OS) and progression-free survival (PFS) in high-risk patients. Independent prognostic analysis, receiver operating characteristic (ROC), C-index, and Nomo chart showed that the model could accurately predict the patients' prognosis. Further analysis showed that cuproptosis associated lncRNAs might participate in the progression of LUSC by affecting citrate-tricarboxylic acid cycling and immune-related functions, and high-risk patients had a greater potential for immune escape. Conclusions In this study, a risk and prognosis model of LUSC cuproptosis associated lncRNAs is constructed. It can accurately predict prognosis and is closely related to multiple LUSC functions.

摘要:

目的 基于癌症基因组图谱(TCGA)数据库建立肺鳞癌的铜死亡相关长链非编码RNAs(long non-coding RNAs,lncRNAs)风险预后模型。方法 基因及临床数据获取自TCGA数据库,铜死亡相关基因获取自铜死亡相关研究。通过共表达分析及Cox回归分析筛选lncRNAs构建预后模型。评估模型的预后价值并分析铜死亡相关lncRNAs与肺鳞癌的关系。组间差异采用Student’s t检验和Wilcoxon检验,Kaplan-Meier生存分析以Log-Rank法检验。结果 共5个lncRNAs参与构建模型。验证分析发现,高风险组患者总体生存率(OS)和无进展生存率(PFS)均较差。独立预后分析、受试者工作特征曲线(ROC)、C-指数和诺莫图显示,该模型可准确预测肺鳞癌患者的预后。进一步分析显示,铜死亡相关lncRNAs可能通过影响柠檬酸-三羧酸循环及免疫相关功能参与肺鳞癌的进展,且高风险患者免疫逃逸的潜能更大。结论 本研究构建了肺鳞癌铜死亡相关lncRNAs风险预后模型,可准确预测肺鳞癌患者预后,并与肺鳞癌多种功能关系密切。
