Expression of gC1qR in sepsis and its effects on macrophage pyroptosis and mitochondrial function in patients with sepsis

gC1qR在败血症中的表达及对败血症患者巨噬细胞焦亡和线粒体功能改变的影响

Abstract:

Objective To investigate the expression of globular C1q receptor (gC1qR) in sepsis and its effects on macrophage pyroptosis and mitochondrial function in patients with sepsis. Methods From January 2020 to May 2022, 60 patients with sepsis in The Second Affiliated Hospital of Nanjing Medical University were selected as an observation group and 60 healthy people as a control group. In the observation group, there were 30 males and 30 females, aged (49.21±18.54) years; in the control group, there were 32 males and 28 females, aged (47.23±20.51) years. The expression of gC1qR in serum was detected by real-time fluorescent quantitative PCR (RT-qPCR). The macrophages were isolated from the patients' whole blood, and were divided into an empty transfection group and a transfection group. The effects of gC1qR on macrophage pyroptosis and mitochondrial function change in patients with sepsis were analyzed. Independent sample t test, paired t test, χ2 test, and Pearson correlation analysis were used for statistical analysis. Results The expressions of serum gC1qR mRNA in the control group and the observation group were (1.00±0.00) and (2.31±0.21) (t=48.320,P<0.001). The pyroptosis rates of macrophages in the empty transfection group and the transfection group were (41.36±5.21)% and (15.44±2.11)%, respectively, with a statistically significant difference between the two groups (t=11.300, P<0.001). The adenosine triphosphate (ATP) contents of macrophages in the empty transfection group and the transfection group were (8.24±2.56) μmol/L and (38.47±4.89) μmol/L, respectively, with a statistically significant difference between the two groups (t=13.420, P<0.001). gC1qR was positively correlated with macrophage pyroptosis (r=0.879, P<0.001), and was negatively correlated with ATP (r=-0.957, P<0.001). Conclusions gC1qR is highly expressed in patients with sepsis. gC1qR can promote the macrophage pyroptosis and lead to the mitochondrial dysfunction.

摘要:

目的 探讨球状C1q受体(gC1qR)在败血症中的表达及对败血症患者巨噬细胞焦亡和线粒体功能改变的影响。方法 选取20201月至20225月南京医科大学附属第二医院的败血症患者60例为观察组,另选取60例健康体检者为对照组。观察组男30例,女30例,年龄(49.21±18.54)岁;对照组男32例,女28例,年龄(47.23±20.51)岁。实时荧光定量聚合酶链反应(RT-qPCR)检测血清中gC1qR表达,同时从患者全血中分离巨噬细胞,将其分化为空转染组和转染组,分析gC1qR对败血症患者巨噬细胞焦亡和线粒体功能改变的影响。统计学方法采用独立样本t检验、配对t检验、χ2检验、Pearson相关性分析。结果 观察组、对照组血清gC1qR mRNA表达分别为(2.31±0.21)、(1.00±0.00),差异有统计学意义(t=48.320P<0.001);空转染组、转染组巨噬细胞焦亡率分别为(41.36±5.21%、(15.44±2.11%,两组比较差异有统计学意义(t=11.300P<0.001);空转染组、转染组巨噬细胞三磷酸腺苷(ATP)含量分别为(8.24±2.56μmol/L、(38.47±4.89μmol/L,两组比较差异有统计学意义(t=13.420P<0.001);gC1qR与巨噬细胞焦亡呈正相关(r=0.879P<0.001)、与ATP呈负相关(r=-0.957P<0.001)。结论 gC1qR在败血症患者中呈高表达,gC1qR可促进巨噬细胞焦亡,并导致线粒体功能紊乱。
